{"id":2330,"date":"2016-09-23T11:34:29","date_gmt":"2016-09-23T11:34:29","guid":{"rendered":"http:\/\/werkstatt.fuelthemes.net\/werkstatt-text-style\/?p=245"},"modified":"2023-04-29T22:22:15","modified_gmt":"2023-04-29T22:22:15","slug":"we-speak-to-ace-tate-creative-fund-board-members","status":"publish","type":"post","link":"https:\/\/www.ronniestanglermd.com\/index.php\/2016\/09\/23\/we-speak-to-ace-tate-creative-fund-board-members\/","title":{"rendered":"Age vs DNA: How Humans Age"},"content":{"rendered":"<p class=\"css-0\"><strong>Our genes may not have as much influence on our aging process as previously believed.<\/strong><\/p>\n<div>\n<article class=\"article-body css-d2znx6 undefined\">\n<div class=\"css-1u22pos\">\n<p>A theory regarding evolution proposes that natural selection is less of a factor for people past their reproductive years since gene variations are not carried forward in offspring at that point in life.&nbsp;A new study &nbsp;in <a class=\"content-link css-1pg8eb5\" href=\"https:\/\/www.nature.com\/articles\/s41467-022-33509-0\" target=\"_blank\" rel=\"noopener noreferrer\">Nature Communications<\/a>.supports this idea, finding that certain tissue types in older people are influenced more by aging itself and by environmental factors than by DNA.&nbsp;These tissues, while critically important, can also lead to cancers, suggesting a limit to what evolution can achieve.<\/p>\n<p>In 1952, Nobel-prize winner <a class=\"content-link css-1pg8eb5\" href=\"https:\/\/www.nobelprize.org\/prizes\/medicine\/1960\/medawar\/biographical\/\" target=\"_blank\" rel=\"noopener noreferrer\">Dr. Peter Medawar<\/a> put forward the hypothesis that aging processes may be a result of evolution\u2019s natural selection not having that much to say about people past their child-bearing years.&nbsp;A new study finds fresh support for <a class=\"content-link css-1pg8eb5\" href=\"http:\/\/www.programmed-aging.org\/theories\/medawar_hypothesis.html\" target=\"_blank\" rel=\"noopener noreferrer\">Medawar\u2019s hypothesis<\/a> in an analysis of how roughly 20,000 human genes are expressed<a class=\"content-link css-1pg8eb5\" href=\"https:\/\/www.genome.gov\/genetics-glossary\/Gene-Expression\" target=\"_blank\" rel=\"noopener noreferrer\">&nbsp;<\/a>as we age.<\/p>\n<p><strong>The study suggests that our genes are less of an influence as we get older.<\/strong><\/p>\n<p>Study senior author <a class=\"content-link css-1pg8eb5\" href=\"https:\/\/ib.berkeley.edu\/people\/faculty\/sudmantp\" target=\"_blank\" rel=\"noopener noreferrer\">Dr. Peter Sudmant<\/a>, assistant professor in integrative biology at the University of California Berkeley has commented:&nbsp;\u201cAlmost all human common diseases are diseases of aging: Alzheimer\u2019s, cancers, heart disease, diabetes &#8230;&nbsp;Massive amounts of public resources have gone into identifying genetic variants that predispose you to these diseases. What our study is showing is that, well, actually, as you get older, genes kind of matter less for your gene expression,\u201d says Sudmant.<\/p>\n<p><span style=\"font-size: revert;\">Dr. Sudmant summarized Medawar\u2019s hypothesis:&nbsp;\u201cGenes that are turned on when we are young are more constrained by evolution because they are critical to making sure we survive to reproduce, while genes expressed after we reach reproductive age are under less evolutionary pressure.\u201d<\/span><\/p>\n<div class=\"css-0\">\n<p><a class=\"content-link css-1pg8eb5\" href=\"https:\/\/www.unical.it\/storage\/teachers\/gAAAAABiPhxOHHbNyMlphAAgu4yM_IJ-uaDHzeWk844XO5qH6gTscT8xRFhEfHcXcMV_ATVA5YWpHWsLjReBHN6BbCWO92hDBg==\/?lang=en\" target=\"_blank\" rel=\"noopener noreferrer\">Dr. Giuseppe Passarino<\/a>, professor of genetics at the University of Calabria in Italy, who was not involved in the study, has explained how this works:&nbsp;\u201cIt is evident that in order to have more children, you need to survive and to be fit [long enough to] reproduce yourself. To get this goal, you need to have no diseases while you are young, to be able to find food, to get a partner. <span style=\"font-size: revert;\">Genes which are expressed during the first part of your lifetime are highly selected, and then <\/span><span style=\"font-size: revert;\">only the&nbsp;<\/span><span style=\"font-size: revert;\">best ones survive.\u201d<\/span><\/p>\n<p>\u201cEvolution is based on the fact that individuals who have better fitness have more children. Thus, their genotypes will spread in the population more than those of subjects who have [fewer] children,\u201d Dr. Passarino added.<\/p>\n<div class=\"css-0\">\n<p><strong>Five Problematic Tissues<\/strong><\/p>\n<p>The researchers retrieved gene expression data for 27 different types of body tissues in almost 950 people from the <a class=\"content-link css-1pg8eb5\" href=\"https:\/\/gtexportal.org\/home\/\" target=\"_blank\" rel=\"noopener noreferrer\">GTEx web portal<\/a>. Individuals were categorized as \u201cyoung\u201d if they were less than 55 years of age, and \u201cold\u201d if they were 55 or over.&nbsp;In their analysis, the researchers found that genetics exerts about the same amount of influence over gene expression in almost all of our tissues until we cross into the \u201cold\u201d bracket.<\/p>\n<p><strong>At that point, aging plays a much more influential role for five critical tissue types &#8211; blood, colon, arteries, esophagus, and fat tissues &#8211; than does genetics.<\/strong><\/p>\n<p>As an influence on gene expression in the study, \u201caging\u201d refers to the universal, progressive cellular aging processes associated with advancing years.<\/p>\n<p>\u201cIn our study, we found in five high proliferation tissues (blood, colon, etc.), [that] these highly constrained genes are actually turned on late in life. These genes tend to be those that are involved in cell division and proliferation, and consequently, in cancer,\u201d said Dr. Peter Sudmant.<\/p>\n<div class=\"css-0\">\n<p><strong>How our environment affects our aging<\/strong><\/p>\n<p>While it would theoretically be helpful if evolution would help select genes that keep us healthy even after we reproduce, according to Dr. Sudmant:&nbsp;\u201cThe \u2018limit of evolution\u2019 here is that, late in life, you really should not have these sorts of genes turned on, and having them turned on actually makes you susceptible to cancer. However, because these are cell types in your body that need to keep turning over \u2014 blood! \u2014 there is no other option.\u201d<\/p>\n<p><strong>Hence, aging and environmental factors are more influential in gene expression for these critical tissues.<\/strong><\/p>\n<p>In the study, environmental influences include factors not directly associated with those processes: the quality of the air and water we breathe and eat, our diet, and also our level of physical exercise.<\/p>\n<p>The study finds that environmental factors account for about a <a class=\"content-link css-1pg8eb5\" href=\"https:\/\/news.berkeley.edu\/2022\/10\/07\/age-vs-genetics-which-is-more-important-for-how-you-age\/\" target=\"_blank\" rel=\"noopener noreferrer\">third<\/a> of gene expression in older people.<\/p>\n<p>Says Dr. Passarino: \u201cThis [study] does not imply that genetics is not important for aging. There are many studies showing that the similarities between relatives regarding the quality of aging (presence of diseases or inabilities) are very high. In fact, although the genes expressed later in life are not selected, still they are important for our life . . .&nbsp;In other words, we are equipped with highly selected alleles for the first part of our life and with alleles [that] are less selected for the second part. However, in both cases, our phenotype is based on our genes.&#8221;<\/p>\n<p><strong>What this study means<\/strong><\/p>\n<\/div>\n<div class=\"css-0\">\n<p>According to Dr. Passarino, to better understand the complexity of how humans age and to learn how to slow down this process, researchers need to study the genes expressed later in life and improve them.&nbsp;\u201cOne option may be to see how the genetic machinery works in long-lived subjects, and try to modulate the machinery of others accordingly,\u201d said Dr. Passarino.&nbsp;\u201dFor instance, it has been observed that long-lived subjects have limited ability to use proteins or sugar. Thus, we can use a limited amount of proteins and sugar to modulate our organism machinery as if we were equipped with the same genes of long-lived subjects,\u201d he elaborated.<\/p>\n<p>\u201cWhen we do studies to identify the genetics underlying disease, we often end up with many genes that we could potentially target. Our study now quantifies how age impacts the expression of genes in the population. We argue that age-associated genes might be better therapeutic targets than the ones that vary in their expression as a function of human genetics,\u201d Dr. Sudmant said.
